ChromoTrax ’s human DNA FISH probes are constructed from microdissected normal human metaphase chromosomes, then directly amplified by PCR using a degenerate primer. PCR labeling reactions have been performed using Fluorescein, Rhodamine or Texas Red. As a control, a sample from every lot has been tested as FISH probes. To pass quality control, the probe must provide good (bright) consistent signal with clean background.Probe DNA are kept in TE buffer.
ChromoTrax's DNA probes are packaged in 10 assay volumes with Hybridization buffers. A volume of 10µl of probe is recommended for each reaction targeting on 22mm x 22 mm slide area/slide. Lower probe concentrations may result in weak or no signals. Multi-color probes require custom manufacture. (For details on experimental steps, please see our attached protocol.)
